High-Throughput Sequencing of Plasma MicroRNA in Chronic Fatigue Syndrome/Myalgic Encephalomyelitis

Abstract: BackgroundMicroRNAs (miRNAs) are known to regulate many biological processes and their dysregulation has been associated with a variety of diseases including Chronic Fatigue Syndrome/Myalgic Encephalomyelitis (CFS/ME). The recent discovery of stable and reproducible miRNA in plasma has raised the possibility that circulating miRNAs may serve as novel diagnostic markers. The objective of this study was to determine the role of plasma miRNA in CFS/ME.ResultsUsing Illumina high-throughput sequencing we identified… Show more

“…Researchers and clinicians seem to agree on the urgent need for unbiased, specific diagnostic biomarkers for ME/ CFS to expedite patient diagnosis and treatment, and to abolish disease stigmas for good 45 . There is also a consensus on the advantages that miRNA profiles from different blood fractions, EVs included, exhibit as molecular diagnostic candidates, and yet, a paucity of studies focusing on ME/CFS miRNA profiling is detected 22,[24][25][26][27] .…”

“…Although, miRNAs such as miR‐150 and miR‐143 were reported in older patients with CFS, especially higher expression and sequencing of plasma miRNAs in patients with CFS/myalgic encephalomyelitis, it is not known whether the proposed miRNAs are expressed or have a role in adolescence with CFS. Therefore, in this study, we investigated the differential expression of 5 miRNAs previously evaluated in older patients with CFS, using whole blood collected from adolescents with CFS and from HCs. Our results show that the expression of miR‐558, miR‐146a, miR‐150, miR‐124, and miR‐143 was significantly downregulated in adolescents with CFS compared with that in HCs.…”

“…This miR, however, has been reported to be upregulated in plasma of CFS/ME patients [19]. It should be pointed out that the differences found across different pain models suggest the existence of disorder specific miRNAs rather than common miRNA regulators of nociceptive modulation.…”
