High throughput T‐DNA insertion mutagenesis in rice: a first step towards in silico reverse genetics

Abstract: SummaryA library of 29 482 T-DNA enhancer trap lines has been generated in rice cv. Nipponbare. The regions flanking the T-DNA left border from the first 12 707 primary transformants were systematically isolated by adapter anchor PCR and sequenced. A survey of the 7480 genomic sequences larger than 30 bp (average length 250 bp), representing 56.4% of the total readable sequences and matching the rice bacterial artificial chromosome/ phage artificial chromosome (BAC/PAC) sequences assembled in pseudomolecules a… Show more

“…The rice mutant serf1 (ssp japonica cv Nipponbare) was obtained from the Génoplante Oryza Tag Line collection (T-DNA insert DAH3F06, line AHHA04) (Sallaud et al, 2004). Homozygous T2 plants were confirmed by PCR using primers 59-AAAACTGATGGCAAATTCGC-39 and 59-GTCT-GGACCGATGGCTGTGTAGAAG-39 for mutant allele and primers 59-GAGTGAGGAGCTCATTGTTTACGA-39 and 59-GTGTCCATGATTCCT-TGGTTTAGT-39 for wild-type allele detection, respectively.…”

“…Multiple efforts are under way to generate a large collection of tagged insertion mutants in rice using T-DNA tagging (Sallaud et al, 2004), Ac/Ds (Greco et al, 2001;Kim et al, 2004), and Tos17 (Miyao et al, 2003). We downloaded from the dbGSS division of GenBank a total of 26,725 sequences reported to be flanking sequence tags (FSTs) from various projects.…”

“…To accelerate the functional characterization of wheat genes, Sallaud et al (2004) proposed using T-DNA/transposon insertion mutants of the orthologous rice genes. However, rice and wheat orthologues do not always have identical functions.…”

“…In Arabidopsis and rice, large gene knockout collections have been assembled from chemical mutagenesis and T-DNA or transposon insertional mutagenesis (Jeon et al 2000;Alonso et al 2003;Sallaud et al 2004). In wheat, a powerful reverse genetics approach was recently implemented through the combination of EMS-mediated mutagenesis and TILLING technology (Slade et al 2005).…”
