Higher‐order oligomerization promotes localization of SPOP to liquid nuclear speckles

Abstract: Membrane‐less organelles in cells are large, dynamic protein/protein or protein/RNA assemblies that have been reported in some cases to have liquid droplet properties. However, the molecular interactions underlying the recruitment of components are not well understood. Herein, we study how the ability to form higher‐order assemblies influences the recruitment of the speckle‐type POZ protein (SPOP) to nuclear speckles. SPOP, a cullin‐3‐RING ubiquitin ligase (CRL3) substrate adaptor, self‐associates into higher‐… Show more

“…Nuclear speckles, also known as interchromatin granule clusters, are membrane-less nuclear organelles enriched in pre-mRNA splicing and transcription factors (Wansink et al, 1993) and recently found to have dynamic, liquid-like properties (Marzahn et al, 2016). Cajal bodies are another type of membrane-less nuclear organelle, enriched in snRNPs and serving as a site of spliceosome assembly (Machyna et al, 2015).…”

“…For example, G3BP1 contains a folded dimerization domain sufficient for SG targeting [79]. Other examples include components of PML and Cajal bodies, and nuclear speckles [8082]. TDP-43 can phase separate by dimerizing via a transient alpha helix in its LCD [11], and can multimerize through its folded N terminal domain [83] (Figure 2B).…”
