Highly efficient control of iron‐containing nitrile hydratases by stoichiometric amounts of nitric oxide and light

Abstract: The reaction of two iron-containing nitrile hydratases (NHase) with NO has been studied: NHase from Rhodococcus sp. R312, which is probably similar to the photosensitive N771 NHase, and the new NHase from Comamonas testosteroni NI1 whose aminoacid sequence is quite different from those of BR312 and N771 NHases. Both enzymes are equally inactivated after addition of stoichiometric amounts of NO added as an anaerobic solution or produced in situ under physiological conditions by a rat brain NO-synthase. Both enz… Show more

“…NHases contain a nonheme Fe 3ϩ or non-corrin Co 3ϩ catalytic center. Iron-type NHases show unique photoreactivity; the enzyme is inactivated by nitrosylation in the dark and immediately reactivated by photo-induced denitrosylation (3)(4)(5). The protein structure is highly conserved among all known NHases (6 -9) as well as a related enzyme, thiocyanate hydrolase (10).…”

“…Fe-type NHase and not Co-type NHase has been shown to be reversibly inactivated in the dark [15] due to the association of an endogenous NO molecule to the iron. The NO molecule is released upon exposure to light, thereby restoring the fully active enzyme, which has been shown for Fe-type NHase from Rhodococcus species N-771 as well as NI1 from Comamonas testosteroni [16][17][18][19]. The 1.7 Å structure of Rhodococcus N-771 Fe-type NHase (which is identical to the R312 enzyme) in its NO inactivated form, reveals an unusual active site [20].…”

“…N771, N774 and R312 and Comamonas testosteroni N11 are photosensitive (Endo and Watanabe 1989;Mayaux et al 1990;Nagamune et al 1990;Bonnet et al 1997). In vivo the enzymes of Rhodococcus sp.…”

“…N771, N774 and R312 can be inactivated by the binding of nitric oxide to the active centre in dark incubated cells and are activated by the photoinduced release of the nitric oxide. The nitrile hydratase of C. testosteroni N11 is not sensitive to dark/NO inactivation in vivo but the purified enzyme is sensitive to inactivation by NO and is subject to photoinduced activation (Bonnet et al 1997). The photosensitivity of the strain AJ270 nitrile hydratase was examined in a number of ways.…”
