Highly sensitive and specific detection of histamine via the formation of a self-assembled magic number cluster with thymine by mass spectrometry

Abstract: A novel method for the detection of histamine (HIM) via the formation of a self-assembled magic number cluster with thymine (T) by electrospray ionization tandem mass spectrometry (ESI-MS/MS) is described. The formation of the magic number cluster [T17 + HIM + 2H](2+) shifts the MS signal of histamine to the interference-free higher mass range and the signal intensity is increased by four orders of magnitude. In addition, the formation of [T17 + HIM + 2H](2+) is highly specific to histamine compared with its m… Show more

“…However, if the host‐guest binding is selective and supramolecular structures survive the electrospray ionization (ESI) process, leading to stable gas‐phase complexes, the resulting ESI‐MS or ESI‐MS/MS signals can be used for analytical purposes. This concept has been applied to direct injection (infusion) analysis of small molecules using metal phthalocyanins, low molecular weight explosives and carboxylic acids using cyclodextrins, and drugs by following the formation of dimers and clusters …”

“…Direct injection methods show however limitations, mainly in samples having high complexity and containing non‐volatile compounds, such as salts. Extraction and/or fractionating procedures might be necessary before injection . Distilled drinks, such as spirits, contain mostly volatile compounds, making the detection of polyamines in these matrices potentially achievable by ESI‐MS with minimum sample processing.…”

“…Amino acids are generally regarded as the essential fundamental units for proteins and polypeptides which participate in a variety of biological matter, such as in cellular functions, regulation, and metabolism, which are significant for the growth and development of living activity. , In addition to being a vital amino acid in many biological systems, histidine (His) is a neurotransmitter and plays an essential role in control of the transportation of some metal ions. Histamine manufactured from His is responsible for a series of physiological processes. Meanwhile, the fluctuation of His’ content is an essential index to reflect variety of potential diseases. For example, when taking in excessive His proteins, the intake will cause an important influence on advanced liver cirrhosis.…”
