Rheumatoid arthritis (RA) is a common chronic autoimmune inflammatory disease, and its etiology is closely related to the overproduction of hypochlorous acid (HClO). However, early detection of RA using an activatable near-infrared-II (NIR-II, 1000−1700 nm) fluorescent probe remains challenging. Herein, we first report an "OFF− ON" NIR-II fluorescent probe named PTA (phenothiazine triphenylamine) for imaging HClO in deep-seated early RA. Electron-rich phenothiazine in the core of PTA was utilized as both an HClOrecognition moiety and a precursor of electron acceptors, displaying a typical donor−acceptor−donor structure with excellent NIR-II emission at 936/1237 nm once reacted with HClO. The probe PTA exhibited good water solubility, high photostability, and rapid response capability toward HClO within 30 s. Moreover, it was able to sensitively and specifically detect exogenous and endogenous HClO in living cells in both visible and NIR-II windows. Notably, PTA enabled the sensitive and rapid visualization of HClO generation in an inflammatory RA mouse model, showing a 4.3-fold higher NIR-II fluorescence intensity than that in normal hindlimb joints. These results demonstrate that PTA holds great promise as a robust platform for diagnosis of HOCl-mediated inflammatory disorders.

Conventional biomedical imaging modalities, including endoscopy, X-rays, and magnetic resonance, are invasive and cannot provide sufficient spatial and temporal resolutions for regular imaging of gastrointestinal (GI) tract to guide prognosis and therapy of GI diseases.Here we report a non-invasive method for optical imaging of GI tract. It is based on a new type of lanthanide-doped nanocrystal with near-infrared (NIR) excitation at 980 nm and second NIR window (NIR-IIb) (1500~1700 nm) fluorescence emission at around 1530 nm. The rational design and controlled synthesis of nanocrystals with high brightness have led to an absolute quantum yield (QY) up to 48.6%. Further benefitting from the minimized scattering through the NIR-IIb window, we enhanced the spatial resolution by 3 times compared with the other NIR-IIa (1000~1500 nm) contract agents for GI tract imaging. The approach also led to a high temporal resolution of 8 frames per second, so that the moment of mice intestinal peristalsis happened in one minute can be captured. Furthermore, with a light-sheet imaging system, we demonstrated a three-dimensional (3D) imaging of the stereoscopic structure of the GI tract.Moreover, we successfully translate these advances to diagnose inflammatory bowel disease (IBD) in a pre-clinical model of mice colitis.

Misplaced or excessive hypochlorous acid in lysosomes has close association with lots of diseases, so monitoring hypochlorous acid in lysosomes is particularly necessary. In this paper, a novel lysosome-targetable fluorescent probe (Lyso-R-HClO) for hypochlorous acid based on HClO-mediated cyclization reaction was developed. In the fluorescent probe, the morpholine unit and the site of HClO-mediated cyclization reaction were respectively used as the lysosome-targetable group and the response group. The probe has a high selectivity and a high sensitivity to hypochlorous acid, with a linear range from 5.0 × 10 -8 to 3.0 × 10 -6 M and a detection limit of 15 nM, and was successfully used to image endogenous and exogenous lysosomal HClO. Finally, Lyso-R-HClO was further applied to image lysosomal HClO produced in bacteria-infected macrophage with satisfactory results, which indicate that it is a useful tool for studies of lysosomal HClO and role of lysosome.
