His‐Tag‐Epitope Imprinted Thermoresponsive Magnetic Nanoparticles for Recognition and Separation Thyroid Peroxidase Antigens from Whole Blood Samples

Abstract: In the epitope imprinting method, a short peptide, even without a defined tertiary structure, can be used as the template to produce protein-selective polymers. In this study, His-tag-epitope imprinted magnetic nanoparticles (EMIP-MNPs) were prepared via epitope imprinting method onto the surface of the magnetic nanoparticles for specific recognition and separation of thyroid peroxidase antigens (TPO). 3-(Trimethoxysilyl) propyl methacrylate (TMSPM), copper (II) acrylate (Cu(II) Ac) and N-isopropylacrylamide (… Show more

“…Tag for protein purification -His tag epitope on the surface of the magnetic nanoparticles -His-tag-epitope imprinted magnetic nanoparticles -Specific recognition and separation of thyroid peroxidase antigens [63] as Ni 2+ , that are coordinated to an EDTA moiety immobilized on a Fe3O4@SiO2 particle [45], or the FLAG-tag [46]. A further kind of directional immobilization relies on exploiting boronic acid moieties or its derivatives, as these would provide anchoring functions to fix the sugar moieties of glycoproteins, through reversible covalent bonds.…”

“…[168] "Epitope" Imprinting: Epitope imprinting are introduced in detail in the abovementioned protein imprinting. [169,170] Owing to amphiphilic and high molecular weight of poly saccharides, it is difficult to prepare MIPs by using the entire poly saccharides as a template. Therefore, similar method, "epitope" imprinting, was introduced into polysaccharides imprinting.…”
