Histamine H3 Antagonists as Wake-Promoting and Pro-Cognitive Agents

Abstract: The histamine H3 receptor is a pre-synaptic auto- and hetero-receptor that controls the release of histamine and a variety of other neurotransmitters in the brain. As such, modulation of the histamine H(3) receptor is expected to affect wake via control of the release of histamine and to affect cognition via regulation of several other neurotransmitters including acetylcholine and norepinephrine. Over the last several years numerous pre-clinical studies have shown that histamine H3 antagonists promote wakefuln… Show more

“…Antagonists and inverse agonists of the H3 histamine receptor subtype promote histamine release and have the potential to improve cognitive performance in neurodegenerative conditions Bacciottini et al 2001;Witkin and Nelson 2004;Arrang et al 2007) potentially without response tachyphylaxis typically associated with agonists. Since histamine exerts its CNS alerting effect, in part, by activating acetylcholine neurons Esbenshade et al 2008;Stocking and Letavic 2008;Passani et al 2001), the effectiveness of a histamine H3 receptor antagonist or inverse agonist may be enhanced by combined use with acetylcholinesterase inhibitors.…”

“…Mutual interactions with these other transmitter systems form a network that links basic homeostatic and higher brain functions, including sleep-wake regulation, circadian and feeding rhythms, immunity, attention, learning, and memory (3)(4)(5)(6)(7)(8)(9). H3 receptor IAs may thus be efficacious in the treatment of various CNS disorders (10,11).…”
