Histidase function in human epidermal cells

Abstract: The activity of histidase was studied in (1) epidermal tissue scraped from human infant foreskin, ( 2 ) fibroblast-like cells in monolayer serial culture from human foreskin, and (3) epithelial-like (epidermal) outgrowth from foreskin primary explants. Foreskin epidermal tissue without in vitro culture and epidermal outgrowth in primary culture from explants of foreskin showed equivalent mean levels of histidase activity, 5.22 x IcMoles urocanic acid produced per milligram protein per minute. Under the same as… Show more

“…The enzyme histidase found in normal epidermis enhances cellular concentration of histidine that can be used for stimulated synthesis of histidine-rich proteins involved in the keratin formation during tumorigenesis (45). Histidase activity in skin carcinoma bearing mouse has been found to be absent and lowered in TPA-treated mice (45,46). Animals showing squamous cell carcinoma following AO/TPA or SANG/TPA administration caused inhibition of histidase activity, which was found to be enhanced following topical application of TOCO and NAC, implicating the decreased synthesis of keratin as observed in histopathological studies.…”

“…Galjaard et al (1974) isolated under a microscope cells or clones grown on a thin plastic sheet but in most laboratories the prevalent morphological cell type (epithelioid or fibroblast-like) is established and enzyme assays are done on mixed cells or in one of the cell types, mainly fibroblast-like cells. Some enzyme activities are expressed exclusively in a particular cell type such as histidase (Barnhisel et al, 1970;Melanqon et al, 1971) and D-amino acid oxidase (Gilbert and Migeon, 1977) in epithelioid cells, and cystathionine synthetase (Uhlendorf and Mudd, 1968;Gerbie et al, 1972;Mudd and Levy, 1978) in fibroblast-like cells. Thus, large variations of enzyme activities were observed in cultured amniotic fluid cells even under carefully controlled culture conditions (Butterworth et al, 1973;Sutherland et al, 1974b;Heukels-Dully and Niermeijer, 1976).…”
