Histologic evaluation of effect of thiocyanate topical application on hamster cheek pouch

Abstract: The present study was conducted to investigate the effect of topical thiocyanate (SCN(-)) application on hamster cheek pouch mucosa. Sixty-six hamsters were divided into two groups. The test substance containing potassium thiocyanate was applied on the hamsters' cheek pouches at 55 mg/kg dosages. Cheek pouches were grossly examined after 12 weeks. Histometric evaluation included height measurements from the epithelium without stratum corneum, stratum corneum, and full epithelium. Clear cells frequency was comp… Show more

“…The sample size was calculated based on the formula for the comparison of means with a 95% significance level, 80% test power, standard deviation of 3.76 μm and a minimum detectable difference between groups of 0.8 μm (Salum et al., ), determining 170 histological fields for investigation per side of the tongue in each group, totalling a minimum of 680 fields. The golden hamsters were randomly distributed in two experimental groups: Group 1—cachaça 48° GL and Group 2—whisky 40° GL.…”

“…We classified the dysplasia according to the presence of these morphological alterations: loss of polarity of basal cells, basilar hyperplasia, increasing nuclear/cytoplasm ratio, epithelial projections resembling tear drops, irregular stratifications of the epithelium, atypical or increased mitosis, presence of mitosis in superficial half of the epithelium, hyperchromatic nuclei, pleomorphic cells, loss of cell cohesion, enlargement of nucleoli, and keratinization in the spinous layer (individual or as a cell group). Presence of up to two of the abovementioned changes represented "mild", three to four represented "moderate" and more than five represented "severe" dysplasia [16]. In the present study, variations in the mucosa of all hamsters were investigated and recorded every week, and histopathological changes were evaluated at 5 and 10 weeks.…”
