This study aims to evaluate the acute toxicity of pymetrozine to juvenile Procambarus clarkii. Two 96-h toxicity tests were conducted to assess the lethal concentration 50 (LC50) values, behaviors, and histopathology (at 50% of the 96 h LC50) after pymetrozine exposure. The results showed high toxicity of pymetrozine to juvenile P. clarkii in a dose and time dependent manner, with a decreasing LC50 from 1.034 mg/L at 24 h to 0.479 mg/L at 96 h. The maximum allowable concentration (MAC) of pymetrozine for P. clarkii was 0.106 mg/L. Behavioral abnormalities were observed in pymetrozine-treated crayfish, such as incunabular hyperexcitability, subsequent disequilibrium, lethargy, and increased defecation. Significant lesions were observed in all pymetrozine-treated tissues, including: (1) in gill, hemocytic infiltration and 33.27% of epithelial cells lesions; (2) in perigastric organs, 64.37%, 29.06%, and 13.99% of tubules with lumen atrophy, vacuolation, and cell lysis, respectively; (3) in heart, 2.5%, 8.55% and 7.74% of hemocytic infiltration, vacuolization, and hyperplasia, respectively; (4) in stomach, 80.82%, 17.77%, 6.98%, 5.24% of cuticula swelling, vacuolization, muscle fragmentation, hemocytic infiltration, respectively; (5) in midgut, 7.45%, 10.98%, 6.74%, and 13.6% of hyperplasia, tissue lysis and vacuolation, hemocytic infiltration, muscle fracture; and (6) in abdominal muscle, 14.09% of myofiber fracture and lysis. This research demonstrates that pymetrozine is highly toxic to juvenile P. clarkii, with significant effects on mortality, behavior and histopathology at concentrations of ≤1.1 mg/L, while the estimated practical concentration of pymetrozine in rice-crayfish culture water was around 20 times lower than the calculated MAC.

Meta-analysis of antiviral protection of white spot syndrome virus vaccine to the shrimp

Currently, white spot syndrome virus (WSSV) is one of the most serious pathogens that impacts shrimp farming around the world. A WSSV vaccine provides a significant protective benefit to the host shrimp. Although various types of vaccines against WSSV have emerged, the immune effects among them were not compared, and it remains unclear which type of vaccine has the strongest protective effect. Meanwhile, due to the lack of effective routes of administration and immunization programs, WSSV vaccines have been greatly limited in the actual shrimp farming. To answer these questions, this study conducted a comprehensive meta-analysis over dozens of studies and compared all types WSSV vaccines, which include sub-unit protein vaccines, whole virus inactivated vaccines, DNA vaccines and RNA-based vaccines. The results showed that the RNA-based vaccine had the highest protection rate over the other three types of vaccines. Among the various sub-unit protein vaccines, VP26 vaccine had the best protective effects than other sub-unit protein vaccines. Moreover, this study demonstrated that vaccines expressed in eukaryotic hosts had higher protection rates than that of prokaryotic systems. Among the three immunization modes (oral administration, immersion and injection) used in monovalent protein vaccines, oral administration had the highest protection rate. In natural conditions, shrimp are mostly infected by the virus orally. These results provide a guide for exploration of a novel WSSV vaccine and help facilitate the application of WSSV vaccines in shrimp farming.

VP28 is an essential WSSV envelope protein used to develop innate immune recognition in shrimp against WSSV, owing to its remarkable capability to localize on the surface of shrimp epithelial cells. Transgenically engineered viral VP28 protein from several expression systems was used to increase the survival chances of shrimp against WSSV pathogen, and their efficacy was found to last for 2-3 weeks following therapy. Though few reports have shown that DNA vaccination using the VP28 DNA construct is capable of protecting shrimp from WSSV, the exact mechanism behind the immunity is still unknown. Several research groups have provided insights into VP28-driven dsRNA vaccination to protect shrimp against WSSV infection, but the stability attained by many of these vaccines turned out to be short-lived. RNAi treatment with VP28dsRNA, shrimp showed better resistance to the WSSV challenge, although multiple doses of dsRNA are required to provide extended protection for up to 28 days. However, no foolproof protection was currently offered by VP28-based recombinant protein or DNA vaccination against WSSV. In recent years, attention has been directed towards identifying small molecule inhibitors of VP28 through molecular docking and molecular dynamics simulation studies and is an emerging approach in drug development and design against WSSV. This review highlights the opportunities, limits and challenges of VP28-based control strategies of White Spot Syndrome Virus, by emphasizing the status quo of current methodological approaches and suggesting future research directions of using it as a potential drug discovery target.
