This study sought to chart the ontogenesis of the goat rumen by histomorphometric examination, scanning electron microscopy and immunohistochemical analysis. A total of 140 goat embryos and fetuses were used, from the first stage of prenatal life until birth. The appearance of the rumen from the primitive gastric tube was observed at 35 days of prenatal life (CRL 3 cm, 23% gestation). By 38 days (CRL 4.3 cm CRL, 25% gestation) the ruminal wall comprised three layers: an internal epithelial layer, a middle layer of pluripotential blastemic tissue and an external layer or serosa. Ruminal pillars were visible at 46 days (CRL 6 cm, 30% gestation), and by 76 days (CRL 18 cm, 50% gestation) ruminal papillae were starting to appear. Under scanning electron microscopy, by 50 days (CRL 7.7 cm, 33% gestation) small ruminal papillae were observed protruding from the surface. Finally, neuroendocrine cells (synaptophysin, SYP) were detected at 53 days (CRL 9 cm CRL, 35% gestation), while glial cell markers (glial fibrillary acidic protein-GFAP, and vimentin-VIM) were found at 108 days (CRL 31 cm, 72% gestation) and 39 days (CRL 4.4 cm, 26% gestation), respectively. Neuropeptide Y (NPY) and vasoactive intestinal polypeptide (VIP) were detected immunohistochemically at 113 days (CRL 33 cm, 75% gestation) and 120 days (CRL 35 cm, 80% gestation), respectively. There are two phases of fore-stomach development during the first 200 days of pouch life in tammar wallaby. For the first 170 days, the mucosa displays an immature gastric glandular phenotype that changes to a cardia glandular phenotype, which remains for the rest of the animal's life. During this 200-day period after birth, the pouch young (PY) is dependent on maternal milk, which progressively changes in composition. We showed previously that PY cross-fostered to host mothers at a later stage of lactation accelerated development. In this study, we investigated whether cross-fostering and exposure to late lactation stage milk affected the transition to cardia glandular phenotype. In fostered PY fore-stomach, there was increased apoptosis, but no change in cell proliferation. The parietal cell population was significantly reduced, and expression of gastric glandular phenotype marker genes (ATP4A, GKN2, GHRL and NDRG2) was down-regulated, suggesting down-regulation of gastric phenotype in fostered PY fore-stomach. The expression of cardia glandular phenotype genes (MUC4, KRT20, CSTB, ITLN2 and LPLUNC1) was not changed in fostered PY. These data suggest that fore-stomach maturation proceeds via two temporally distinct processes: down-regulation of gastric glandular phenotype and initiation of cardia glandular phenotype. In fostered PY, these two processes appear uncoupled, as gastric glandular phenotype was down-regulated but cardia glandular phenotype was not initiated. We propose that milk from later stages of lactation and/or herbage consumed by the PY may play independent roles in regulating these two processes.

The study was conducted on 36 developing reticulum from healthy and normal embryos/ foeti of Indian goat. Embryos/foeti were assigned into three groups. Histo-differentiation of reticulum of goat stomach took place at 38 days of gestation. The wall of foetal reticulum was made up of three strata i. e. epithelium, pleuripotent blastemic tissue and serosa up to 49 days of fetal age and definite four layers viz. epithelium, propria-submucosa, tunica muscularis and serosa were observed first at 51 days of gestation. Upto 100 days of gestation the reticulum was lined by undifferentiated stratified epithelium and thereafter gradually transformed into stratified squamous epithelium. Keratinization was noticed at term. Primary and secondary reticular crests made their appearance at 60 and 112 days of gestation, respectively. Corial papillae were detected in primary and secondary reticular crests at 121 and 145 days of gestation, respectively. Near term, the core of the reticular crest also contained muscularis mucosae in its proximal 1/3rd region while core of the corial papillae contained only lamina propria. Reticular, collagen and elastic fibers came into sight at 46, 100 and 134 days of gestation, respectively.Summary statementThe study was conducted on intrauterine reticulum of Indian goat. From this study it can be concluded that the histogenesis of reticulum was almost completed in prenatal life. However, to become functional it still required more time as the relative sizes of reticulum and process of keratinization were yet to be completed.
