Histone H3K9 demethylase JMJ25 epigenetically modulates anthocyanin biosynthesis in poplar

Abstract: Anthocyanins are involved in several aspects of development and defence in poplar (Populus spp.). Although, over the past decades, significant progress has been made in uncovering these anthocyanin biosynthetic and regulatory mechanisms, the fundamental understanding of the epigenetic regulation in this pathway is still largely unclear. Here, we isolated a histone H3K9 demethylase gene JMJ25 from Populus and characterized its role in anthocyanin biosynthesis by genetic and biochemical approaches. JMJ25 was ind… Show more

“…In poplar, few studies on gene editing by CRISPR/Cas9 have been published in the last four to five years [22,23,30,50,51,52,53]. All studies have in common that CRISPR/Cas9 has successfully been applied to insert a mutation in one or more genes for knockout (KO) of the poplar target gene.…”

“…[53] knocked out all NST/SND orthologs and confirmed their central role in the secondary cell wall (SCW) formation in wood fibres, xylem ray parenchyma cells and phloem fibers in hybrid aspen. [50] discovered an epigenetic mechanism that modified anthocyanin biosynthesis in poplar. Following knockout of a histone H3K9 demethylase gene JMJ25 , MYB182 expression is altered leading to a change of the histone methylation status in the chromatin.…”

In rice, JMJ706 regulates transcription factor expression required for spikelet development, floral morphology and organ number (Sun and Zhou, 2008) Arabidopsis JMJ16 represses leaf senescence (Liu et al, 2019) and JMJ25 functions in poplar to remove H3K9me2 marks and modulate anthocyanin biosynthesis (Fan et al, 2018).…”

“…For example, DNA methylation of the MdMYB1 promoter region affects the transcript levels of MdMYB1 and determines anthocyanin content in skin of two apple cultivars (Ma et al , 2018). Also, a histone H3K9 demethylase, JMJ25, directly affects the transcript levels of MYB182 , an anthocyanin repressor, to modulate anthocyanin biosynthesis in poplar (Fan et al , 2018). In morning glory, flower color variation can be caused by transposon insertions or excisions into anthocyanin biosynthetic or regulatory genes (Inagaki et al , 1994; Hoshino et al , 2001; Morita et al , 2014, 2015).…”
