Histone lysine demethylases as targets for anticancer therapy

Abstract: It has recently been demonstrated that the genes controlling the epigenetic programmes that are required for maintaining chromatin structure and cell identity include genes that drive human cancer. This observation has led to an increased awareness of chromatin-associated proteins as potentially interesting drug targets. The successful introduction of DNA methylation and histone deacetylase (HDAC) inhibitors for the treatment of specific subtypes of cancer has paved the way for the use of epigenetic therapy. H… Show more

“…The ATL-specific coherent relationship comprising the epigenetic "writer" and "eraser" may contribute to locking or stabilizing the epigenetic states. 58 We also approached the regulatory mechanisms of EZH2 abundance and genomic occupancy. A series of experiments in normal T cells and in ATL models demonstrated the necessity of a complicated signaling network for sufficient EZH2 induction.…”

“…This is largely due to the requirement for chromatin to be reorganized to create access to key differentiation loci. The opening of chromatin is aided by the recruitment of histone acetyl transferases (HATs), chromatin remodelling enzymes and histone methyltransferases (HMTs), as well as the recently discovered histone lysine demethylases (KDMs) and ten-eleven translocation (TET; also known as methylcytosine dioxygenase) proteins that initiate DNA demethylation [130][131][132][133][134][135][136] (FIG. 5b).…”

“…In contrast, inhibition of histone demethylases would shift the balance between methylation and demethylation towards higher methylated histones. Histone (de)methylating enzymes are also specific, with several dozen specimens in several families described to date (reviewed in Højfeldt et al, 2013). So far, however, there are no reports of active demethylation processes in insects.…”
