Histones of Chlamydomonas reinhardtii (Synthesis, Acetylation, and Methylation)

Abstract: Histones of the green alga Chlamydomonas reinhardfii were prepared by a new method and fractionated by reversed-phase high-performance liquid chromatography. Acid-urea-Triton gel analysis and tritiated acetate labeling demonstrated high levels of steady-state acetylation for the single histone H3 protein, in contrast to low levels on histones H4 and H2B. Twenty percent of histone H3 is subject to dynamic acetylation with, on average, three acetylated lysine residues per protein molecule. Histone synthesis in l… Show more

“…However, using antibodies specific for H3K9me2 or H4K20me2, we were unable to detect a signal corresponding to histones H3 or H4, respectively (data not shown). This is in agreement with previous data indicating that Chlamydomonas may not contain significant amounts of dimethyl H3K9 (Waterborg et al, 1995). When we examined the global levels of H3K9me1, all strains behaved similarly to the wild type ( Figure 4D).…”

“…Under normal Chlamydomonas growth conditions, 81.2% of histone H3 is monomethylated at K4, and 15.6% shows K9 monomethylation (Waterborg et al, 1995). Consistent with our ChIP results, the extensive amount of H3K4me1 (equivalent to the silenced chromatin fraction) strongly indicates that this modification is associated with repressed chromatin.…”

“…In some eukaryotes including C. reinhardtii, monomethylation of lysine 4 in histone H3 (H3K4) and monomethylation of H3K9 are putative epigenetic markers for a transcriptionally silent/inactive chromatin state (Cheung and Lau 2005;Van Dijk et al 2005;Waterborg et al 1995;Casas-Mollano et al 2007;Pfluger and Wagner 2007), and the underlying DNA in heterochromatin is often accompanied by methylation. In contrast, histones located in euchromatic regions usually have different types of modifications, such as acetylation, and the DNA in these regions is usually not highly methylated.…”

“…Proteins were quantitated by densitometry of gels, stained with Coomassie Brilliant Blue R-250, and destained in 7% acetic acid, 20% methanol. Specific radioactivity was determined by densitometry of fluorographic exposures exactly as described (17,18).…”
