Historical background of umbilical stem cell culture

Abstract: Umbilical cord is a waste material, and therefore does not raise ethical concerns related to its use for research and medicine. Stem cells from umbilical cord have a significant advantage over cells from other sources. First, the umbilical cord is an infinite source of stem cells, because it can be taken theoretically during each delivery. Secondly, acquisition of umbilical cord is a non-invasive, safe procedure for mother and child. Thirdly, the transplantation of umbilical cord stem cells is associated with … Show more

“…It was performed in France in a child with Fanconi Anaemia (FA) (Ballen et al 2013 ). In Poland, the first cord blood stem cell transplant took place in 1994 to treat a boy suffering from acute myelogenous leukaemia (Borys-Wójcik et al 2019 ). In 2007, there was the first Polish transplant of umbilical cord blood which was previously collected, processed and stored in a private bank (Borys-Wójcik et al 2019 ; PBKM 2007 ).…”

Since then, umbilical cord blood stem cells have been transplanted over 40.000 times worldwide including a few transplants in Poland (Ballen 2017 ; FamiCord Group 2020 ).…”

“…The first recipient of umbilical cord blood stem cells was a 5-year old boy with severe Fanconi anemia in 1988 [ 16 ]. In Poland, a successful frozen and banked umbilical stem cell transplantation in a boy with acute myeloblastic leukemia was performed eight years later [ 17 ]. Initially, UBC transplantation was limited to children, and both related and unrelated cord blood transplants have been performed with high rates of success for a variety of hematologic disorders and metabolic storage diseases [ 16 ].…”

“…Then, the collected samples are directly placed in culture flasks (explant cultures) or enzymatically digested (collagenase I/II, hyaluronidase, trypsin). In the next step, the processed solution is filtered or centrifuged and transferred into culture dishes and cultured in various media at 37 °C in a 5% CO 2 atmosphere [ 197 , 223 , 224 , 225 ]. Fortunately, the isolation of MSCs from the umbilical cord tissue is much more efficient.…”
