HIV-1 Glycoprotein 41 Ectodomain Induces Activation of the CD74 Protein-mediated Extracellular Signal-regulated Kinase/Mitogen-activated Protein Kinase Pathway to Enhance Viral Infection

Abstract: Besides mediating the viral entry process, the human immunodeficiency virus (HIV-1) envelope protein gp41 can bind to many host cell components and regulate cell functions. Using a yeast two-hybrid system, we screened a human bone marrow cDNA library and identified a novel gp41-binding protein, CD74 (the MHC class II-associated invariant chain). Here, we report possible biological effects mediated by interaction between gp41 and CD74. We found that HIV-1 gp41 could bind directly to host CD74 in HIV-1-infected … Show more

“…[52] Antigen presentation antagonism HIV-1 gp41 interacts with HLA-associated invariant chain and may have a role in MHC-directed antagonism. [28,53] doi:10.1371/journal.ppat.1003258.t001 , but nine proteins are produced due to editing events in the GP gene. The GP gene primary transcript encodes for a ,110 kDa, dimeric secreted GP (pre-sGP).…”

“…When peripheral blood mononuclear cells (PBMCs) were incubated with recombinant post-fusion HIV-1 gp41, an increase in phosphorylated ERK occurred. Furthermore, this activation was inhibited in a dose-dependent manner by treatment with soluble recombinant CD74 ectodomain [28]. The activation of the ERK/MAPK pathway via high levels of the CC-chemokine RANTES (or other exogenous signals) is responsible for increased infectivity of HIV-1 [29].…”

“…Several polymorphisms have been identified in both the GSTO1 and GSTO2 genes that could have an effect on this capability. Deletion of Glu155 results in decreased stability and activity of the GSTO1 protein product (Whitbread et al, 2003; Schmuck et al, 2008; Zhou et al, 2011). The Glu155 deletion is estimated to be carried by 3% of Europeans and may be as high as 10% in Chinese populations (Whitbread et al, 2003).…”

“…gp120 plays a role in the immunostimulatory effects related to HIV-1-associated dementia ( 21 ). gp41 has been shown to significantly enhance HIV-1 infection and replication in vitro ( 22 ). Likewise p24 and p17 have been shown to possess potent immunostimulatory properties leading to increased HIV-1 replication in PBMCs of infected individuals receiving cART ( 23 ), as well as activated PBMCs in vitro ( 16 ).…”
