HL-A Antigens and Genes - III. Production of HL-A Typing Antisera of Desired Specificity

Abstract: Eight skin graft donor-recipient pairs were selected by lymphocyte typing for production of HL-A typing antisera. After the second or third skin graft from the same donor, antibodies appeared in the sera of five of the recipients. The antibodies were directed against antigens which seem to be determined by alleles of the LA and 4 sub-loci, and the specificity showed close correlation to the HL-A incompatibility between donor and recipient. By re-immunization with intradermal leucocyte injections, two additiona… Show more

“…A major incompatibility with respect to unidentified HL-A antigens may then exist. In this way antigen FJH was identi fied, most probably determined by a new allele at the Four sub-locus [16]. The LA antigen HL-A9 (Lcll, LA4, TH -BS) was identified by Walford et al in a similar way [21].…”

“…Typing reagents were obtained by screening thousands of sera from parous women, transfused patients or individuals immunized by planned skin grafting and leukocyte injections [17]. The sera were treated aseptically and stored at -20°C or -75°C or at 4°C after lyophilization.…”
