Homogeneous probing of lipase and α-amylase simultaneously by AIEgens

Abstract: An AIE dual-reactive supramolecular probe has been devised to simultaneously measure endogenous lipase and α-amylase activity in a homogeneous system. Fluorescence quantitative analysis of lipase and α-amylase in real biological samples enables rapid and accurate diagnosis of diseases.

“…36 This indicated that the lipase enzyme was able to diffuse into the film and retained its catalytic activity. Since an abnormally high level of lipase features a condition known as acute pancreatitis, 37 which imposes a severe threat to people's lives, the PNPP-doped film is promising for easy detection of the degree of lipase activity in a clinical setting.…”

“…Taking advantage of the restriction of cavities toward rotors of AIEgens, Tang's group constructed an AIE dual-reactive supramolecular probe based on TPE-modified γ-CD. [106] This supramolecular probe can be used in homogeneous probing of lipase and α-amylase, thereby enabling [8], PEG-Np, and PTPE, and their use as building blocks for the preparation of SNPs; B) schematic diagram of SNPs for application in imaging-guided drug delivery. “…Inflammation induces a significant increase in α-amylase in blood under the conditions of pancreatic and sialaden injury. 37,38 More importantly, α-amylase can hydrolyze α-1,4 glycosidic bonds in carbohydrates, including cyclodextrins (CDs), 39 which provides a stimulus to control the fluorescence of AIEgens-cyclodextrin (CD) based hydrogels. Herein, by taking advantage of the host–guest recognition of TPE with γ-CD, an example of a self-healing hydrogel has been designed and derived from the host–guest interaction between TPE modified poly( l -glutamic acid) (PLGA–TPE) and acrylate γ-CD (Ac-γ-CD) under the assistance of chemical crosslinking.…”
