Homoisoflavanone prevents mast cell activation and allergic responses by inhibition of Syk signaling pathway

Abstract: Homoisoflavanone inhibited mast cell activation through the suppression of Syk pathway together with the inhibition of cPLA2 . Thus, it might be a good candidate molecule for allergic diseases.

“…During degranulation, the secretory granules of mast cells can fuse with plasma membrane using SNAREs, and prestored molecules, such as histamine and β-hexosaminidase, are then released into the extracellular environment [8,43,44]. Thus, the release of β-hexosaminidase is widely used for an indicator of degranulation of mast cells [12,[14][15][16][17]19,23,45]. In this context, the effect of cinnamaldehyde derivatives on degranulation was assessed by β-hexosaminidase assay.…”

“…The effects of IgE and PMA/A23187 on mast cells are known to activate PKC and extracellular Ca 2 + uptake [6,11,23,42,44,46]. These effects control degranulation, cytoskeleton rearrangement, and gene expression of pro-inflammatory cytokines in mast cells [5-8, 19,46].…”

“…BMMCs were generated as previously described, with minor modifications (Frossi et al, 2007;Lee et al, 2014). Briefly, bone marrow cells were obtained from tibias and femurs of 8-12-week-old C57BL/6 mice.…”
