Homologous Overexpression of Diacylglycerol Acyltransferase in Oleaginous Fungus Mucor circinelloides WJ11 Enhances Lipid Accumulation under Static Solid Cultivation

Abstract: Diacylglycerol acyltransferase (DGAT) catalyzes the binding of acyl-CoA to diacylglycerol to form triacylglycerol (TAG). Previous studies strongly indicate that DGAT2, rather than DGAT1, is crucial for TAG accumulation in the oleaginous fungus Mucor circinelloides. To increase the lipid content of M. circinelloides WJ11, McDGAT2 was overexpressed by homologous recombination; compared to the control strain Mc2075, transformants McDGAT2d showed a significant increase in biomass for both spores and mycelia (from … Show more

“…The promotion of DGAT on lipid accumulation has been reported in many previous studies. The homologous overexpression of McDGAT2d gave an improvement to lipid accumulation under static solid fermentation [ 30 ], and the overexpression of endogenous PtDGAT1 increased by more than double in TG and total lipid compared to that of the wild-type Phaeodactylum tricornutum , which reached 57% and 73% of dry weight, respectively [ 31 ]. In addition, the overexpression of endogenous PtDGAT2B resulted in enhancing the neutral lipid content by 69% [ 32 ].…”

“…circinelloides strain WJ11 (wild-type) [6] was used as a reference in this study. M. circinelloides WJ11 defective in the carRP gene (∆carRP WJ11) was employed for investigating lipid and carotenoid biosynthesis, with reference to the Mc2075 strain with knock out of the carRP gene performed through homologous recombination by using the designed pMAT2075 plasmid [28].…”

“…However, the downregulation of acetyl-CoA synthetase (EC: 6.2.1.1) and ATP citrate lyase (EC: 2.3.3.8) might lead to a reduction in the acetyl-CoA pool for fatty acid synthesis [28]. Downregulation of fatty acid synthase (FAS1) (EC: 2.3.1.86), involved in long-chain fatty acid synthesis, was also found in ∆carRP WJ11.…”
