Honokiol Potentiates Pentobarbital-Induced Sleeping Behaviors through GABAAReceptor Cl-Channel Activation

Abstract: − This study was undertaken to investigate whether honokiol could enhance the pentobarbitalinduced sleeping behaviors through γ-aminobutyric acid (GABA) receptor Cl -channel activation. Thirty minutes after the oral administration of honokiol, mice were received sodium pentobarbital (42 mg/kg, i.p.). The time elapsed from pentobarbital injection to the loss of the righting reflex was taken as sleeping latency. The time elapsed between the loss and voluntary recovery of the righting reflex was considered as the… Show more

“…Animals that failed to fall asleep within 15 minutes after pentobarbital administration were excluded from the experiments. [24][25][26] Primary culture of cerebellar neurons Primary cultures of cerebellar neurons enriched in granule cells were prepared from cerebella of 8-day-old male Sprague-Dawley rats as previously reported. 27 After cell culture for 8 days, cells express functional GABA A receptors, with an expression pattern similar to that apparent in the cerebellum during postnatal development but different from that observed in the adult rat cerebellum.…”

“…Additional treatment of primary cultured cerebellar granule cells showed selective GABA A receptor α-subunit expression increase, on the other hand, chronic honokiol treatment did not affect GAD abundance. These results suggest that honokiol potentiates pentobarbital-induced sleep through GABA A receptor Cl - channel activation [ 40 ]. In 2012, Wei-Min Qu et al .…”

“…Pentobarbital enhances the effects of GABA and induces sleep in a predictable fashion ( Ticku and Maksay, 1983 ). Sedative-hypnotics or anti-anxiety agents have also been shown to increase pentobarbital-induced sleep duration ( Ma et al ., 2008 ). Treatment with caffeine, however, suppresses the expression of GABAergic systems and results in sleep disturbances ( Mukhopadhyay and Poddar, 2000 ; Li et al ., 2004 ; Mabunga et al ., 2015 ).…”
