Host succinate is an activation signal for Salmonella virulence during intracellular infection

Abstract: Key to the success of intracellular pathogens is the ability to sense and respond to a changing host cell environment. Macrophages exposed to microbial products undergo metabolic changes that drive inflammatory responses. However, the role of macrophage metabolic reprogramming in bacterial adaptation to the intracellular environment has not been explored. Here, using metabolic profiling and dual RNA sequencing, we show that succinate accumulation in macrophages is sensed by intracellular Salmonella Typhimurium… Show more

“…Mineral M9 contained: i) 12.8 g/l Na2HPO4×7H2O, 3 g/l KH2PO4, 0.5 g/l NaCL, 1 g/l NH4Cl, 2mM MgSO4, 0.1 mM CaCl2, 0.0004% Histidine, 0.5 g/ml Thiamine; ii) trace elements: 134 M EDTA, 31 M FeCl3×6H2O, 6.2 M ZnCl2, 0.76 M CuCl2×2H2O, 0.42 M CoCl2×2H2O, 1.62 M H3BO3, 81 nM MnCl2×4H2O; and iii) supplemented with 40 mM Na-succinate (Sigma-Aldrich) as carbon source. Bacterial cultures were inoculated to an OD600 nm 0.01 and incubated at 37°C without aeration for microaerobic conditions as previously described (Rosenberg et al, 2021;Serna et al, 2010) or with 200 rpm shaking for aerobic conditions. For solid growth, Salmonella strains were streaked on LB or M9 Agar supplemented with 40mM Na-succinate as sole carbon source.…”

“…Overall, METLIN-NL empirically derived data will enable new types of analyses facilitating more rapid identification of unknown compounds via both fragment ion and neutral loss similarity searching. 2 Both biologists and chemists will be able applying METLIN-NL to the structure elucidation of unknowns derived from animals, 19 plants, 14,20 or microbiota 21 ; and METLIN-NL can also be used as a resource for identifying unexpected synthetic chemical or enzymatically modified drug products (e.g. pharmaceuticals 22 ) as it is populated with both biological and chemical entities.…”
