Housing conditions during self-administration determine motivation for cocaine in mice following chronic social defeat stress

Abstract: Rationale: Stress exposure has a lasting impact on motivated behavior and can exacerbate existing vulnerabilities for developing a substance use disorder. Several models have been developed to examine how stressful experiences shape drug-reward. These range from locomotor sensitization and conditioned place preference, to the propensity for drug self-administration or responding to drug-predictive cues. While self-administration studies are considered to have more translational relevance, most of the studies t… Show more

“…Environmental enrichment attenuates yohimbine-induced reinstatement of cocaine seeking following selfadministration in rats (Chauvet et al, 2009). The ability of social defeat stress to promote cocaine seeking in mice is heightened when mice are isolated during self-administration (Engeln et al, 2021).…”

“…Importantly, our mice were pair-housed throughout the fentanyl procedure. Previously (Engeln et al, 2021), we showed that when mice are single-housed, CSDS-susceptibility is associated with increased early cocaine self-administration. When pair-housed, CSDS instead decreases cocaine selfadministration, analogous to increased anhedonia.…”

“…Numerous studies demonstrate psychosocial stress exerts differential effects on drugabuse-like behavior (Neisewander et al, 2012;Newman et al, 2018). Previously, we showed individual stress-response impacts cocaine self-administration (Engeln et al, 2021). To test the hypothesis that susceptibility to psychosocial stress increases homecage opioid consumption, we subjected mice to chronic witness defeat (CWDS) or social defeat stress (CSDS) (Timeline and schematic in Fig 1A ).…”
