How preclinical studies have influenced novel psychoactive substance legislation in the UK and Europe

Abstract: Novel psychoactive substances (NPS) are new drugs of abuse. Over the last 10 years 50–100 new NPS have been detected for the first time each year. This has led to numerous deaths and challenges to healthcare providers and law‐makers worldwide. We review preclinical studies of NPS and discuss how these studies have influenced legislative decisions. We focus on the UK legal system but include experiences from Europe. We reviewed manuscripts from 2008–2019 and have summarised the in vitro and in vivo data on NPS,… Show more

“…• Strong research evidence (e.g. Cochrane reviews, meta-analyses, high-quality randomised controlled trials); 10 In particular, studies on the toxicity and management of the harms of NPS and risks associated with long-term use and dependence liability are few, partly because most NPS have limited or no medical use, and partly because some of these substances have only recently emerged.…”

“…Only a small proportion of NPS have been researched in preclinical studies of which some have influenced changes to the law in U.K. and EU [ 91 ]. One of our studies [ 64 ] is among those as it was used by the ACMD in their recommendation for Class B substance classification for Benzofurans [ 92 ], which led to the Amendments to the Misuse of Drugs Act 1971 in 2014.…”

“…In particular, Lenzi and colleagues have demonstrated the mutagenic capability of synthetic cathinone and cannabinoids [ 39 , 40 ], underlying their potential toxicity. However, in vitro tests cannot easily predict pharmacokinetics or penetration into the human brain [ 41 , 42 ]. Therefore, animal models have played a key role in preclinical research explaining the neurobiological, psychopathological, behavioral and etiological aspects, and studying drug dependence and acute/chronic abuse [ 43 ].…”
