Acute phase proteins (APPs) are highly conserved plasma proteins that are increasingly secreted by the liver in response to a variety of injuries, independently of their location and cause. APPs favor the systemic regulation of defense, coagulation, proteolysis, and tissue repair. Various APPs have been applied as general diagnostic parameters for a long time. Through proteomic techniques, more and more APPs have been discovered to be differentially altered. Since they are not consistently explainable by a stereotypic hepatic expression of sets of APPs, most of these results have unfortunately been neglected or attributed to the nonspecificity of the acute phase reaction. Moreover, it appears that various extrahepatic tissues are also able to express APPs. These extrahepatic APPs show focally specific roles in tissue homeostasis and repair and are released primarily into interstitial and distal fluids. Since these focal proteins might leak into the circulatory system, mixtures of hepatic and extrahepatic APP species can be expected in blood. Hence, a selective alteration of parts of APPs might be expected. There are several hints on multiple molecular forms and fragments of tissue-derived APPs. These differences offer the chance for multiple selective determinations. Thus, specific proteoforms might indeed serve as tissue-specific disease indicators.

In this study, we aimed to investigate the protective effect of Vitamin U (Vit U) on amiodarone (AMD)-induced hepatotoxicity. Male Sprague-Dawley rats were randomly divided into four groups. Group I was control animals receiving corn oil for 7 days, Group II consisted of animals receiving Vit U (50 mg/kg) for 7 days, Group III consisted of animals receiving AMD (100 mg/kg) for 7 days and Group IV consisted of animals given AMD and Vit U (in same dose and time). AMD and Vit U were administered to rats by gavage. On the 8th day, all the animals fasted overnight were sacrificed. Blood and liver tissue samples were taken from animals. Serum total cholesterol levels and liver gamma glutamyl transferase (transpeptidase) adenosine deaminase and tissue factor activities, liver glycoprotein component level such as fucose, hexose and hexosamine were determined. All the parameters were found to be increased in AMD group as compared to control group. Administration of Vit U reversed these effects in AMD group. According to these results, we can conclude that Vit U can prevent AMD-induced liver injury.

INTRODUCTION: Leprosy is a fairly dreaded disease, but it is curable. However, liver failure is one of the side effect of the treatment that challenging to manage.OBJECTIVES: Assessing the effects of Multi Drug Therapy (MDT) on the liver function (SGOT, SGPT) and Mycobacterium Leprae Particle Agglutination (MLPA) test (IgM anti PGL-1) before and after treatment in patients with multibacillary (MB) leprosy.METHOD: Twenty-eight patients who met the inclusion criteria were enrolled in this study which categorized as new MB leprosy patients in Dr. Wahidin Sudirohusodo Hospital, Makassar, Indonesia. In order to test the liver function, blood serum was taken to measure the SGOT and SGPT level with Bochringer Mannheim automatic analysis, while MLPA test measurement was performed with qualitative method. Blood serum was collected three times with the following period; before the treatment, 3 months, and 6 months after treatment. The data was analyzed using Friedman and Wilcoxon Signed Rank test with significant level p<0.001.RESULT: There were significant increases in SGOT and SGPT levels (p<0.001) before and after MDT treatment between 3 to 6 months. Meanwhile, for IgM anti PGL-1, it was not significant (p>0.01) before treatment and after 3 months treatment, but significant different (p<0.001) on 6 months treatment.CONCLUSION: MDT treatment on MB leprosy patient increase the SGOT and SGPT level but decrease the IgM anti PGL-1 after the 6 months of treatment.
