Hsa-miR-155-5p Up-Regulation in Breast Cancer and Its Relevance for Treatment With Poly[ADP-Ribose] Polymerase 1 (PARP-1) Inhibitors

Abstract: miR-155-5p is a well-known oncogenic microRNA, showing frequent overexpression in human malignancies, including breast cancer. Here, we show that high miR-155-5p levels are associated with unfavorable prognostic factors in two independent breast cancer cohorts (CSS cohort, n = 283; and TCGA-BRCA dataset, n = 1,095). Consistently, miR-155-5p results as differentially expressed in the breast cancer subgroups identified by the surrogate molecular classification in the… Show more

“…In breast cancer, the oncomir miR-155-5p was found mostly upregulated and associated with a more aggressive type and worse outcome (122). The combined treatment with Olaparib, an inhibitor of PARP1, with miR-155-5p ectopic overexpression was found a great combination to induce lethal effect to reduce cancer cells viability (123) Further investigation of PARPs as a biomarker for novel cancer therapies is needed.…”

“…Without patient-level data, it would be very difficult to tell whether miR-155 is actually useful as a marker in early detection of BC. Besides, BRCA1-deficient tumors have been shown to have increased levels of miR-155, which could be relevant for identifying possible respondents to PARP-1 inhibitors [ 55 , 56 ]. Most of the mechanistic studies on miR-155 and BC have been done on these patients, which was ignored in this study.…”

“…The top hit in correlated miRNAs, mir−155, is a known oncogenic microRNA that targets the ELK3 transcription factor that is imperative in the response to hypoxia [ 17 ]. It was shown to be upregulated in breast cancer and linked to PARP-1 inhibitors response [ 18 ]. In bladder cancer, this miR is known to be significantly overexpressed, promoting tumour growth by repressing DMTF1, a tumour-supressing gene [ 19 ].…”
