HSV-mediated transfer of interleukin-10 reduces inflammatory pain through modulation of membrane tumor necrosis factor α in spinal cord microglia

Abstract: To dissect the molecular basis of the neuroimmune response associated with the genesis of inflammatory (nociceptive) pain, we constructed a herpes simplex virus-based gene transfer vector to express the antiinflammatory cytokine interleukin-10 (IL-10), and used it to examine the effect of IL-10 expression in activated microglial cells in vitro, and in inflammatory pain in vivo. IL-10 reduced the phosphorylation of p38 mitogen-activated protein kinase (MAPK) and decreased the expression of full-length membrane … Show more

“…[21]. Inoculation of rats with a herpes simplex virus IL-10 vector, 10 days before intraplantar formalin administration, resulted in reduced pain sensitivity and decreased expression of TNF-␣ in the spinal cord [22]. Elevated levels of IL-10 have been suggested to be of pathophysiological significance in autoimmune and inflammatory diseases, such as lupus erythematosus [23], systemic sclerosis [24] and vesicular bullous diseases [25,26].…”

“…We have previously reported that nonreplicating HSV vectors similar to the one used in this study are effective in delivering transgenes to DRG in vivo (49,50), and the transgene product is released by the central afferent terminals of transduced DRG (51). We confirmed that subcutaneous inoculation of QHNgSR resulted in the production of NgSR in DRG neurons in vivo.…”

“…Expression of interleukin-10 (IL-10), an anti-inflammatory cytokine, was reported to reduce pain-related behaviors in the rodent formalin model, an effect likely achieved via the reduction of proinflammatory factors (e.g., tumor necrosis factor [TNF]) in spinal microglia. 10 11 The neuropathic pain observed in trigeminal neuralgia can be modeled in rodents with the ligation of the infraorbital nerve, part of the maxillary division of the trigeminal system. The maxillary nerve is the branch carrying sensory information from the cheek area, or in rodents, the whisker pad.…”
