HuIFN-γ Antiviral Activity Correlates with Immunological Reactivity in a Double-Monoclonal Antibody Radiometrie Assay

Abstract: Several lines of evidence have been obtained that correlate biological activity with reactivity in an immunoradiometric assay (IRMA) employing two different monoclonal antibodies. Antiviral activity and immunological reactivity decreased in parallel when freeze-dried human gamma-interferon (HuIFN-gamma) was heated for prolonged periods of time at elevated temperatures. Likewise, aqueous preparations of HuIFN-gamma inactivated by acid pH or heat responded similarly in the biological assay and IRMA. These result… Show more

“…v-lnterferon was determined in the concentrated BAL fluid of the same 10 patients, in which the alveolar macrophage activity could be estimated, by an immunoradiometric assay using reagents obtained from Centocor [20]. A volume of 0.2 ml of each sample (or control standard material) was incubated in the well of a reaction tray along with polystyrene beads coated with a monoclonal anti body against v-interferon for 2 h at room temperature.…”

“…The gamma interferon concentrations in the specimens were determined with a standard curve. In this test, described by Chang et al (4), the monoclonal antibodies are specific for the epitopes of biological activity of human interferon (23) and can detect titers as low as 0.20 IU/ml.…”
