“…Virus was purified on CsCl gradients and particle counts determined using the equation 1 mg/ml virus protein = 3.45 × 10 12 particles/ml. 28 Infectivity was measured by plaque assay in 293 cells; particle:infectivity ratios of 10-50 were obtained for all virus stocks.…”

“…A cloned version of the novel adenovirus genome was identified, the viral genome was released from the plasmid by restriction enzyme digestion, and virus replication was initiated by transfecting the genome into the E1-complementing 293 cells (24) using a polyethylenimine transfection method (23). Virus was amplified and purified from cell lysates by using CsCl density gradient centrifugation as described (25); virus was quantified from protein content (25) by using the conversion factor 1 mg/ml pure virion protein ϭ 3.4 ϫ 10 12 viral particles/ml (26). Control adenovirus AdJ5 is an Ad5 vector with the same viral genotype (E1/E3 defective) but lacking an expression cassette.…”

“…Quantification of viruses was based on protein content, 29 using the conversion 1 mg of viral protein ¼ 3.4 Â 10 12 viral particles (vp). 30 Typical vector stock titers were 10 12 vp/ml with a 1:100 infectious plaque forming unit to vector particle ratio.…”
