Human CC10, the homologue of rabbit uteroglobin: genomic cloning, chromosomal localization and expression in endometrial cell lines

Abstract: Human and rat cDNAs to Clara CeU 10 kDa protein {CClO) have been previously isolated. Comparison of the amino acid sequences showed that CCIO is homologous to rabbit uteroglobin. Here we present further evidence that human CClO is the human COUnterpartor rabbit uteroglobin. We have isolated the gene and have mapped its genomic Jocalization to chromosome llq11-qter. Sequence anaJysis of the 5' -ßanking region reveals tbat the homology between the human and the rabbit gene starts at the first exon/intron boundar… Show more

“…The use of the 5' region of human ccI OkD gene has been suggested in gene transfer experiments (4). This might elucidate the significance of cis-acting elements for the cell-specific expression of the human gene and might lead to the possible use of portions of its 5' region in gene therapy for obtaining organ-specific expression of a wild-type gene in the respiratory epithelia.…”

“…The name ccI OkD derives from the apparent molecular mass ofthis protein in nonreducing SDS-PAGE (1). The cDNA (3) as well as the gene (4) for this protein have been cloned and characterized. The exact physiological role of cc10kD is yet to be clarified.…”

“…Region I encompasses the noncanonical TATA-box region (TACA), which is recognized by the tissuespecific factor TCF and the ubiquitous factor TPF (27), which was identified in this study as YY1. (B) Comparison of the rabbit uteroglobin (UG) TACA box (56), the cognate human (63), mouse (54), and rat (18) TATA sequences, the TATA box of the Ad2 major late promoter (6), and the TACA box of the Ad2 EIIaL promoter (21) with the TATA consensus sequence, using the weight matrix description of Bucher (6). A negative weight matrix score indicates the extent of deviation from the consensus sequence (score ϭ 0).…”

“…1]). Alignment of the rabbit uteroglobin TATA-box sequence with the corresponding sequences of the human and rat genes (whose gene products were formerly called Clara cell 10-kDa secretory proteins [CC10] [60]) shows that the human and rat genes possess a canonical TATA box, whereas the rabbit promoter has a TACA box with a cytosine instead of a thymine in the third position of the consensus sequence. Though TBP/TFIID has been reported to bind to noncanonical TATA box sequences (47,59), all available data on TACA boxes point to the fact that it is a weak recognition site for TBP (7,24,39).…”
