Human Colon and Liver Cancer Cell Proliferation Inhibition by Peptide Hydrolysates Derived from Heat-Stabilized Defatted Rice Bran

Abstract: Rice bran, an economical, underutilized coproduct of rough rice milling, was used to produce peptide hydrolysates, which were investigated for anticancer activity. Protein hydrolysates prepared by Alcalase hydrolysis under optimized conditions were treated further to obtain gastrointestinal (GI)-resistant peptide hydrolysates. They were fractionated into >50, 10-50, 5-10, and <5 kDa sizes and evaluated for inhibitory activity on proliferation of human colon (Caco-2) and liver (HepG2) cancer cell lines by Trypa… Show more

“…The hydrolysates were treated with simulated gastrointestinal resistant solutions to generate GIE resistant peptides, which were then fractionated into <5, 5-10, 10-50, and >50 kDa fractions as previously done 11 . 2.2.2 Anti-obesity effect: Evaluation of degree of differentiation of adipocytes for accumulation of lipids: Undifferentiated human preadipocytes (Lonza, USA) were allowed to differentiate upon treatment with a known differentiating agent [0.25µmol/L DEX (IS-IBMX-DEX) mixture] (Sigma USA).…”

“…Advanced cancer treatment methods employ a holistic approach by incorporating dietary or nutritional, spiritual and physical therapies along with drugs and radiation. As part of nutritional supplementation, bioactive peptides have a prominent role in alternative medicine which involves controlling diseases using food derived macro molecules (Zhang et al 1998;Korhonen and Pihlanto 2003;Kannan et al 2008). Certain other bioactive molecules are from natural sources like plants (Ng et al 2003;Ohba et al 2004;Silva-Sánchez et al 2008).…”

“…Certain other bioactive molecules are from natural sources like plants (Ng et al 2003;Ohba et al 2004;Silva-Sánchez et al 2008). One of the major approaches to produce bioactive peptides was enzymatic hydrolysis or fermentation, especially from sources like soybean, wheat, corn, rice, sunflower, milk, eggs, and meat (Gibbs et al 2004;Wang and de Mejia 2005;de Mejia and de Lumen 2006;Kannan et al 2008Kannan et al , 2009Kannan et al , 2010Hettiarachchy 2013).…”
