Human Cytomegalovirus Glycoprotein UL16 Causes Intracellular Sequestration of NKG2D Ligands, Protecting Against Natural Killer Cell Cytotoxicity

Abstract: The activating receptor, NKG2D, is expressed on a variety of immune effector cells and recognizes divergent families of major histocompatibility complex (MHC) class I–related ligands, including the MIC and ULBP proteins. Infection, stress, or transformation can induce NKG2D ligand expression, resulting in effector cell activation and killing of the ligand-expressing target cell. The human cytomegalovirus (HCMV) membrane glycoprotein, UL16, binds to three of the five known ligands for human NKG2D. UL16 is retai… Show more

“…58 To draw a conclusion, a large scale and time-course analysis of blood cells of patients with PNH is needed. With regard to the discrete features of ULBP3, 61 our inhibition assays with antibodies indicate that ULBP3 was not engaged in the killing of K562 cells in contrast with ULBPs 1 and 2.…”

“…Proportions of PNH granulocytes in lanes 4 to 7 were 35%, 15%, 1%, and 100%, respectively. Both lack of membrane ULBPs and up-regulation of ULBP2 mRNA in lanes 5 and 7 may be explained by inhibition of their display on the membrane (so-called masking) 58 and the absence of GPI ϩ granulocytes (patient no. 6 in Table 1), respectively.…”

“…The endogenous ligands for human NKG2D are MHC class I chain-related proteins (MIC)A and MICB and another family of class I-related molecules, the UL16-binding proteins (ULBP)1-3. Some NKG2D ligands are up-regulated in cells infected with CMV (47,48), but the signals regulating the expression of MIC and ULBP in the context of other viral infections have yet to be defined.…”

“…Blocking this interaction would therefore be of obvious benefit to the virus as emphasized by the demonstration that deletion of miR-UL112-1 led to an increased susceptibility to NK killing. Interestingly, HCMV also encodes a protein, UL16, which was previously shown to inhibit MICB signaling by intracellular sequestration, suggesting a level of redundancy in targeting of MICB (Dunn et al, 2003).…”
