Human defensins and cytokines in vaginal lavage fluid of women with bacterial vaginosis

Abstract: HBD-2 and HD-5 may be involved in defending against invasion by BV-related microorganisms and the decrease in IL-4 concentration may increase susceptibility to BV.

“…In PMN granules, ␣-defensins attain mg per ml concentrations (30); however, AP concentrations likely decline significantly upon release into the extracellular milieu, where H. ducreyi is most likely to encounter the APs. Estimates of extracellular AP concentrations in human tissues vary extensively among studies, with vaginal concentrations of HNP-1 to -3 ranging from 0.2 g per ml (45) to 3 g per ml (5) and an estimated 60 to 110 g of HD5 per ml (18); urethral concentrations of HD5 were estimated at 0.5 to 18 g per ml (38). The concentration range used in our assays covers most estimates of physiologically relevant doses.…”

“…They are divided into two classes, ␣-and ␤-defensins, based on the pattern of disulfide bonds that stabilize their tertiary structure (1,2). HD5 is one of six human ␣-defensins and is constitutively expressed and secreted in the female and male genitourinary tracts (3)(4)(5). Concentrations of HD5 in vaginal lavage fluid of healthy women have been reported to be 16.5 Ϯ 10.5 M (3).…”

“…Sexually transmitted infections (STIs) increase the risk of HIV mucosal transmission in part by altering epithelial barriers (10,16,24). Individuals with STIs exhibit elevated levels of antimicrobial peptides, including human defensin 5 (HD5) produced by genital epithelial cells and human neutrophil peptides (HNPs) produced by infiltrating neutrophils (9,27,30,35). These defensins are believed to play a role in controlling bacterial infection.…”
