Human glioblastoma arises from subventricular zone cells with low-level driver mutations

Abstract: Glioblastoma (GBM) is a devastating and incurable brain tumour, with a median overall survival of fifteen months. Identifying the cell of origin that harbours mutations that drive GBM could provide a fundamental basis for understanding disease progression and developing new treatments. Given that the accumulation of somatic mutations has been implicated in gliomagenesis, studies have suggested that neural stem cells (NSCs), with their self-renewal and proliferative capacities, in the subventricular zone (SVZ) … Show more

“…It is widely accepted that GBM originate from the stem cell reservoirs responsible for adult neurogenesis in the brain (Singh et al, 2004;Zhu et al, 2005;Siebzehnrubl et al, 2011;Westphal and Lamszus, 2011). There are several reports that support that neural stem cells localized at the subventricular zone (SVZ) are the origin of glioblastoma (Lee et al, 2018;Gimple et al, 2019). Neural stem cell fate is controlled by environmental cues, among which cytokines play a crucial role (Ming and Song, 2011).…”

“…4 In the world each year, there are 100 000 new cases of glioblastoma. 6 The histological grading of gliomas is categorised according to World Health Organization (WHO) guidelines. Strikingly, the 5year relative survival (just 4.6% at 5 years) for glioblastoma patients has remained stable over the last three decades.…”

“…However, recent evidence indicates that glioblastoma arises instead from neural stem cells within the subventricular zone of the brain. 6 The histological grading of gliomas is categorised according to World Health Organization (WHO) guidelines. The highest grade (grade IV) of glioma is glioblastoma, and the diagnosis depends on observations of high cellularity, atypical glial cells, and microvascular proliferation (MVP), or significant necrosis or both.…”

“…In the brain, glial cells are immersed in an electric field created by tissue polarity from brain macrostructures as well as the local field potentials which are established from the action potentials fired by the neurons 12 . A weak endogenous electric current has been shown to serve as a guidance cue for neuroblast migration from the subventricular zone in mouse 13 , a region speculated as the origin of glioma tumorigenesis 14 . Thus, the physiological a) amy.shen@oist.jp electric fields in the brain may play an important role in mediating the glioma tumorigenesis and invasion [15][16][17][18][19] .…”
