“…After washing six times for 5 minutes with the washing buffer, antigen-antibody reaction was detected using an enhanced chemiluminescence (Amersham), followed by exposure to an autoradiography film (Kodak, Tokyo, Japan) for 5 to 30 seconds. A T-cell line, Jurkat, 39 was used as a positive control for bcl-2 expression, and PLC/PRF/5 40 and HepG2 41 were used as other controls as HCC cells. Because the enhanced chemiluminescence method enhances the antigen-antibody reaction signal, the treated and untreated samples were diluted 1:2 and 1:4 to show if the rise in Bcl-2 and p21 WAF-1 are caused by a true induction by butyrate treatment.…”

HepT1 (Pietsch et al, 1996) and HUH6 (Japanese Collection of Research Bioresources, Osaka, Japan) are HB cell lines, HepG2 (ATCC, Manassas, United States) is a cell line of a HB with hepatocellular carcinoma (HCC)-like features (López-Terrada et al, 2009), and HUH7 is a well differentiated HCC cell line (He et al, 1984;Alexander, 1987). All cell lines were cultured in RPMI 1640 (Life Technologies, Carlsbad, CA, United States), supplemented with 10% (v/v) fetal bovine serum (FBS, Life Technologies) and 1% Pen/Strep in a humidified chamber containing 5% CO 2 at 37 °C.…”
