Human Monoclonal IgE Antibodies—a Major Milestone in Allergy

Abstract: Purpose of Review Bound to its high affinity receptor on mast cells and basophils, the IgE antibody molecule plays an integral role in the allergic reaction. Through interactions with the allergen, it provides the sensitivity and specificity parameters for cell activation and mediator release that produce allergic symptoms. Advancements in human hybridoma technologies allow for the generation and molecular definition of naturally occurring allergen-specific human IgE monoclonal antibodies. … Show more

“…IgE also plays a pivotal role in responses to allergens, such as: anaphylactic reactions to drugs, bee stings and antigen preparations used in desensitisation immunotherapy. 4 In allergic conditions, high titres of IgE are observed—and this was also the case in the patient we describe here.…”

“…However, it is also essential for type I hypersensitivity reactions that manifest in various allergic diseases such as allergic asthma and rhinitis, food allergies, chronic urticaria and atopic dermatitis. “…IgE-secreting human hybridomas are subsequently screened for specificity, selected, cloned and expanded. Secreted hIgE mAb, expressed in serum-free medium, are purified via anti-IgE immunoaffinity chromatography ( 3 ). Using this approach, panels of hIgE mAb with naturally occurring pairing of IgE heavy and light chains have been developed (which overcame a limitation of engineered recombinant antibodies).…”

“…It is widely believed that various factors such as insect bites, Epstein-Barr virus, human herpesvirus, and Candida albicans may disrupt T-cell regulation or trigger an IgE-mediated type 1 hypersensitivity reaction. 20 , 21 The presence of increased levels of eosinophils and IgE in the peripheral blood of KD patients suggests that Th1 and Th2 cells, as well as T regulatory cells, may play a role in the development of KD. Ohta et al employed flow cytometry to analyze T-cell subsets and discovered a significantly higher proportion of Th2 cells in KD patients compared to the control group.…”
