Human paraneoplastic antigen Ma2 (PNMA2) forms icosahedral capsids that can be engineered for mRNA delivery

Victoria Madigan,
Yugang Zhang,
Rumya Raghavan
et al.

Abstract: A number of endogenous genes in the human genome encode retroviral gag -like proteins, which were domesticated from ancient retroelements. The paraneoplastic Ma antigen (PNMA) family members encode a gag -like capsid domain, but their ability to assemble as capsids and traffic between cells remains mostly uncharacterized. Here, we systematically investigate human PNMA proteins and find that a number of PNMAs are secreted by human cells. We determine that PNMA2 fo… Show more

“…It has been noted that PNMA capsids should be suitable as delivery vehicles for therapeutic agents such as RNA vaccine delivery and gene therapy 18,19,32,82 ; such therapies involve bioengineering capsid proteins for the delivery of designer cargo, such as mRNA. The beauty of PNMA1 and PNMA4 is that they would require no engineering to function as therapeutic agents for subfertility because they could naturally harbor pro-fertility cargo.…”

“…We reasoned that the conservation of PNMA genes is unlikely to be based on their autoantigenic properties after ectopic expression in neoplasms. While Pnma5 is implicated in mouse oocyte development via an unknown mechanism 17 , little is known about the non-pathological roles of the other genes other than that some seem to have retained the ability to form capsids when expressed in human cells or bacteria 18,19 . The autoantigenicity of PNMA2, in fact, requires capsid formation 20 .…”

“…Obviously, the probabilities for containing more than one electron are infinitesimally small. For comparison, parameters from a typical cryo‐EM experiment, such as for the FSC shown in Figure 1b (Madigan et al, 2024 ), might involve a total fluence of ~30 e − /Å 2 , fractionated into 24 frames, so in each frame there would be an expectation of ~1.25 electrons/Å 2 . It was stated in Jonaid et al that the frames were then motion‐corrected to compensate for the large translations of the virions over the course of the 20 s movies.…”
