Human Parathyroid Hormone: Antibody Characterization

Abstract: PTH antibodies were raised in two sheep (S 469 and S 478) by immunizing with porcine and bovine parathyroid extracts. Both antisera were characterized with various PTH preparations and fragments. Both antisera cross react with human, bovine and porcine PTH, one antiserum also binds rat PTH. Region specificity could be attributed to the mid region of the PTH molecule with particularly high affinities of both antisera for the fragment 44-68 hPTH. S 478 has similarly high affinity for intact hormone (affinity con… Show more

“…The assay system employed i2sI-labelled bovine PTH (Inolex) and sheep antiserum (S478) against porcine and bovine PTH. Serum PTH measurements were only performed in a random sample of the population (n=76) [6]. Urinary excretion of cyclic adenosine monophosphate (CAMP) was determined in duplicate by a radioimmunoassay method with use of a commercial kit (Amersham, UK).…”

“…The reference range was for calcium 2.20-2.60 mmol/l (8.8-10.4 mg/dl), magnesium 0.65-1.00 mmol/l (1.6-2.4 mg/dl), phosphate 0.76-1.44 mmol/I (2.4-4.S mg/dl) and alkaline phosphatases 0.8-4.8 pkat/l. Serum PTH was analyzed before the study and after 4 months using a radioimmunoassay technique detecting intact PTH and the C-terminal 2/3 of the molecule [8],…”

“…Serum iPTH levels were determined by radioimmunoassay with the use of antiserum $478 [12]. Conjunctival and corneal calcification were evaluated by slit lamp and hand light examination at 6 monthly intervals.…”
