Human SRY inhibits β-catenin-mediated transcription

Abstract: In most mammals, sex is determined by the presence or absence of the SRY gene. SRY encodes a DNA binding HMG-box transcription factor which, during embryogenesis, is the initial trigger of testis differentiation from the bipotential gonad, yet its precise mode of function remains unclear. In ovarian development, R-spondin1 and Wnt4 act through the Wnt/β-catenin signaling pathway to regulate TCF-dependent expression of unknown target genes and repress testis development. Conversely, SRY may be necessary to prev… Show more

“…25,26,28 Several transcription partners such as Sox5/6, 46 cMaf, 47 TRAP230, 48 p300, 20 Barx2, 49 Smad3, 21 PIAS, 50 b-catenin, 51 and Scleraxis 33 can modify Sox9-dependent transcription in chondrogenesis and sex determination. Co-activator p300 acts as a bridging factor for connecting DNA-binding transcription factors to the transcriptional apparatus and as a protein scaffold to form multicomponent transcriptional complexes.…”

“…Several transcription partners such as Sox5/6 (Ikeda et al, 2004;Lefebvre et al, 2001), Barx2 (Meech et al, 2005), β-catenin (Akiyama et al, 2004;Bernard et al, 2008), c-Maf (Huang et al, 2002), PIAS (Hattori et al, 2006), and TRAP230 (Zhou et al, 2002) can modify the Sox9-dependent transcription in chondrogenesis and sex determination. Sox9 is necessary for mesenchymal condensations, and considered to be a key transcription factor for the following expression of Sox5/6 genes (Akiyama et al, 2002).…”

“…In XY gonads, WNT/ b-catenin signaling is downregulated following Sry expression. Consistent with this observation, in vitro studies have shown that SRY can antagonize CTNNB1 by physically interacting with it at the protein level, thus targeting CTNNB1 to nuclear bodies, to trigger its degradation and inhibit CTNNB1-mediated transcriptional activity (Bernard et al 2008). Nevertheless, this mechanism has not yet been confirmed in vivo during sex determination.…”
