Hydration shell differentiates folded and disordered states of a Trp-cage miniprotein, allowing characterization of structural heterogeneity by wide-line NMR measurements

Abstract: Hydration properties of folded and unfolded/disordered miniproteins were monitored in frozen solutions by wide-line 1 H-NMR. The amount of mobile water as function of T (−80 °C < T < 0 °C) was found characteristically different for folded (TC5b), semi-folded (pH < 3, TCb5(H+)) and disordered (TC5b_N1R) variants. Comparing results of wide-line 1 H-NMR and molecular dynamics simulations we found that both the amount of mo… Show more

“…This is the basis of the approach to determine the connectivity of water molecules via intramolecular vibrations [67,68]. There is now no doubt that a change in the conformation of a protein is accompanied by a change in its water shell [69], and this is inextricably linked to protein functioning [70]. The reverse is also true: a change in the water shells leads to a change in the conformation of proteins [15].…”

“…This method allows studying of mobility of various nuclei, which is a huge advantage. Moreover, NMR spectroscopy allows one to characterize the local environment of a particular nucleus and to characterize not only translational but also rotational mobility [ 74 , 75 , 76 , 77 ]. This makes it an indispensable tool for studying mechanisms of transfer processes, which can include different stages.…”

“…The most notable feature is the presence of prominent peaks around 1630 cm −1 that can be assigned to β-sheet structures. Since in bulk phase no β-structure is detectable (Taricska et al, 2019), the appearance of βsheet and β-turn structures and the decreased presence of random coil and α-structures points to surface induced structural changes.…”
