Hydroxychloroquine attenuates autoimmune hepatitis by suppressing the interaction of GRK2 with PI3K in T lymphocytes

Abstract: Hydroxychloroquine (HCQ) is derivative of the heterocyclic aromatic compound quinoline, which has been used for the treatment of autoimmune diseases. The central purpose of this study was to investigate therapeutic effects and inflammatory immunological molecular mechanism of HCQ in experimental autoimmune hepatitis (AIH). Treatment with HCQ ameliorated hepatic pathologic damage, inflammatory infiltration, while promoted regulatory T cell (Treg) and down-regulated CD8+T cell differentiation in AIH mice induced… Show more

“…Research has shown that HCQ can rebalance the immune response mediated by Th17/Treg and improve SLE [ 11 ]. HCQ primarily affects T cell lipid metabolism, thereby promoting the differentiation and function of Tregs [ 12 ]. Additionally, HCQ can inhibit the proliferation of T cells and B cells, leading to reduced production of T follicular helper (Tfh)-related cytokines CXCL13, type I and type II interferons, and immunoglobulins [ 13 ].…”

“…Again, thanks to the study by Beck et al [ 106 ] and the study resumed by Jin et al [ 115 ], Hydroxychloroquine would appear to have some effect on the CCR4 receptor, probably due to its quinoline ring nitrogen, which may be able to interact with the receptor pocket [ 116 ].…”

“…HCQ is known to work by inhibiting the release of pro-inflammatory cytokines, including IFN, TNF, and IL-12, increasing the release of anti-inflammatory cytokines like TGF-β1 and by promoting Treg differentiation and activity (Jin et al, 2022). HCQ is also known to restore the Th17/Treg balance, leading to significantly lower expression of IL-17 in Th17 cells and higher levels of Foxp3 and TGF-β (An et al, 2017).…”
