Hydroxyl carlactone derivatives are predominant strigolactones inArabidopsis

Abstract: ABSTRACTStrigolactones (SLs) regulate important aspects of plant growth and stress responses. Many diverse types of SL occur in plants, but a complete picture of biosynthesis remains unclear. In Arabidopsis thaliana, we have demonstrated that MAX1, a cytochrome P450 monooxygenase, converts carlactone (CL) into carlactonoic acid (CLA), and that LBO, a 2-oxoglutarate-dependent dioxygenase, converts methyl carlactonoate (MeCLA) into a metabolite called [… Show more

“…The LGS1 gene encodes a sulfotransferase‐like protein (Gobena et al ., 2017). Since sulfotransferases generally transfer a sulfo group to a hydroxyl group of their substrates (Hirschmann et al ., 2014), it seemed possible that the substrates of LGS1 are hydroxylated CL and hydroxylated CLA (Yoneyama et al ., 2020). We also noted that 4DO, a stereoisomer of 5DS, is synthesized via 18‐OH‐CLA in rice (Fig.…”

“…The noncanonical SL biosynthetic gene LBO is present in many seed plant species and acts in the later steps after CLA synthesis (Yoneyama et al ., 2020). It is a novel branching‐associated gene originally identified in Arabidopsis (Brewer et al ., 2016).…”

“…Arabidopsis LATERAL BRANCHING OXIDOREDUCTASE ( LBO ) encodes a 2‐oxoglutarate‐dependent dioxygenase that oxidizes MeCLA to 1′‐HO‐MeCLA to act during the final steps of noncanonical SLs biosynthesis (Brewer et al ., 2016). Although most MeCLA is converted to CLA by LBO in vitro , 1′‐HO‐MeCLA is indeed present as an endogenous compound in Arabidopsis (Yoneyama et al ., 2020). Moreover, CL and MeCLA can accumulate in the roots of Arabidopsis lbo mutants, and the morphological characterizations induced by MeCLA accumulation are intermediate between the wild‐type Ws‐4 and the max4 mutant plants.…”

“…Recently, the direct conversion of carlactonoic acid to orobanchol without passing through 4-deoxyorobanchol has been described ( Wakabayashi et al., 2019 ). Moreover, a cytochrome P450 and a 2-oxoglutarate-dependent dioxygenase genes were identified being involved in SL synthesis in Lotus japonicus ( Mori et al., 2020 ), and hydroxyl carlactone derivatives as relevant intermediaries in SL synthesis have been identified in Arabidopsis ( Yoneyama et al., 2020 ). Despite the active research, our knowledge about the details of SL biosynthesis after carlactone is still limited ( Bouwmeester et al., 2019 ).…”
