Hydroxytyrosol mildly improve cognitive function independent of APP processing in APP/PS1 mice

Abstract: These results suggest that HT may represent as a functional ingredient in Mediterranean diet in ameliorating AD-involved neuronal impairment via modulating mitochondrial oxidative stress, neuronal inflammation, and apoptosis without affecting APP processing.

“…Although HT mildly benefited cognitive behaviour in APP/PS1 mice, it had no effect on brain Aβ accumulation. In addition, HT was associated with amended mitochondrial dysfunction, increased SOD‐2 expression, and reduced brain inflammatory markers (Y. Peng et al, ).…”

“…Although many biological effects have been investigated, it has not been shown whether and how HT acts efficiently in AD progression. HT administered to 3‐month‐old female transgenic APP/PS1 mice at 5 mg/kg/day for 6 months resulted in electroencephalographic activity and cognitive function improvements and reductions in mitochondrial oxidative stress and neuroinflammation (Peng et al, ). In the same study, HT not only successfully rescued altered mitochondrial complex IV expression but also normalized mitochondrial complex I–IV activities compromised in this transgenic AD brain.…”

“…Additionally, GSH depletion and HO‐1 and NQO‐1 upregulation observed in AD mice in response to Aβ‐induced oxidative stress were reversed by HT. Remarkably, APP/PS1 mice showed NF‐κB/MAPK‐dependent activation of NLRP3 inflammasome with increased IL‐6 and COX‐2 expression, which were significantly decreased by HT administration (Peng et al, ).…”

“…Among the minor constituents of EVOO, a number of polyphenols and secoiridoids, including oleocanthal, hydroxytyrosol and oleuropein aglycone, are able to modulate many pathways that are relevant to the aging process. The beneficial effects of some components of EVOO in preventing and delaying the onset of AD, and in reducing the severity of its symptoms, have been found in transgenic mouse models 40,41 . On the other hand, the population cohort study, Nurses' Health Study, showed that greater adherence to the Mediterranean diet was associated with longer telomeres 42 .…”
