Hypercapnia downregulates hypoxia‐induced lysyl oxidase expression in pulmonary artery smooth muscle cells via inhibiting transforming growth factor β1 signalling

Abstract: Hypoxic pulmonary arterial hypertension is characterized by elevated pulmonary vascular resistance and remodelling. Transforming growth factor-β 1 (TGF-β 1 ) is the master regulator in cellular response to hypoxia which can directly target lysyl oxidase (LOX). This study aimed to determine whether hypercapnia attenuates hypoxic pulmonary hypertension via regulating TGF-β 1 and LOX signalling. We found that exposure to hypercapnia ameliorated the increase in mean pulmonary artery pressure (mPAP) and ratio of ri… Show more

“…TGF-β is a central determinant of the cellular response to hypoxia. Coordinated upregulation of LOX and TGF-β, both at the protein and mRNA levels, have been shown to be induced in vitro and in vivo by hypoxia, and downregulated by hypercapnia in a rat model of hypoxic pulmonary hypertension [ 42 ].…”

“…The active substances in black ginseng can affect the occurrence of inflammatory response by regulating the TLRs signaling pathway ( Figure 1). Ginsenoside Rb1 can inhibit the protein level expression of TLR3 and TRIF mRNA, and then inhibit the activation of the TLR3/TRIF signaling pathway, thereby reducing the inflammatory lung response in asthmatic rats [31] …”

“…Ginsenoside Rb1 can inhibit the protein level expression of TLR3 and TRIF mRNA, and then inhibit the activation of the TLR3/TRIF signaling pathway, thereby reducing the inflammatory lung response in asthmatic rats. [31] Ginsenoside Rg1 can inhibit the activation of NF-кB and the phosphorylation of transforming growth factor β-activated kinase 1 in macrophages stimulated by LPS, and effectively inhibit the combination of LPS and TLR4. In vivo, oral administration of ginsenoside Rg1 has slowed down TNBS-induced colonic shortening.…”
