Hyphal Tip-Associated Localization of Cdc42 Is F-Actin Dependent in Candida albicans

Abstract: The rho-type GTPase Cdc42 is important for the establishment and maintenance of eukaryotic cell polarity. To examine whether Cdc42 is regulated during the yeast-to-hypha transition in Candida albicans, we constructed a green fluorescence protein (GFP)-Cdc42 fusion under the ACT1 promoter and observed its localization in live C. albicans cells. As in Saccharomyces cerevisiae, GFP-Cdc42 was observed around the entire periphery of the cell. In yeast-form cells of C. albicans, it clustered to the tips and sides of… Show more

“…This analogy cannot be taken too far, as proteins such as Far1p that are critical for shmoo formation in the budding yeast are not required for germ tube development in C. albicans (5). However, the ability to initiate polarized growth from a nonconstricted point of emergence, the ability to migrate the nucleus into this polarized extension, and the requirement for the actin cytoskeleton to direct the localization of the Cdc42p GTPase (52) are common characteristics of shmooing yeast cells and C. albicans cells initiating a germ tube.…”

“…Conditions that disrupt proper cell cycle progression can result in polarized growth through diverse cell cycle pathways (Fig. 4) (20,33,219). Notably, depending on the phase of cell cycle arrest, the cells will have different filamentation phenotypes.…”

“…Cortical actin patches are clustered at the hyphal tips during the hyphal growth phase (Wendland, 2001). In the ectomycorrhizal basidiomycete Suillus bovinus and the dimorphic fungus Candida albicans during hyphal growth stages Cdc42p was found to be localized to hyphal tips, indicating that Cdc42p is not only required for hyphal tip formation but might also play a role during polarized hyphal growth (Gorfer et al, 2001;Hazan and Liu, 2002).…”
