Hypoallergenic Variants of the Major Latex Allergen Hev b 6.01 Retaining Human T Lymphocyte Reactivity

Abstract: Hev b 6.01 is a major allergen of natural rubber latex with sensitization of 70–86% of latex glove-allergic subjects. Recently, we mapped the immunodominant T cell sites of Hev b 6.01 to the highly IgE-reactive hevein (Hev b 6.02) domain. “…A reliable blood test to predict or confirm allergy to Gelofusine would enable a simpler and safer management strategy for individuals with possible Gelofusine allergy. The flow cytometric basophil activation test (BAT), with detection of surface expression of CD63 (a lysosomal membrane glycoprotein) on activated basophils, has been useful in identifying subjects allergic to neuromuscular blocking drugs [9] and beta-lactams [10] and more recently we have used this assay to confirm hypoallergenic latex allergens [11]. We report a case series where peri-operative anaphylaxis due to Gelofusine was confirmed by skin testing and BAT, suggesting that the BAT is a reliable screening test for Gelofusine allergy in such patients, avoiding the risks associated with skin testing.…”
