Hypoxia Alters the Response to Anti-EGFR Therapy by Regulating EGFR Expression and Downstream Signaling in a DNA Methylation–Specific and HIF-Dependent Manner

Abstract: Intratumoral hypoxia occurs in 90% of solid tumors and is associated with a poor prognosis for patients. Cancer cells respond to hypoxic microenvironments by activating the transcription factors, hypoxia-inducible factor 1 (HIF1) and HIF2. Here, we studied the unique gene expression patterns of 31 different breast cancer cell lines exposed to hypoxic conditions. The EGFR, a member of the ErbB (avian erythroblastosis oncogene B) family of receptors that play a role in cell proliferation, invasion, metastasis, a… Show more

“…Most CGIs in gene promoters are unmethylated, allowing active transcription [ 5 ]. CGI methylation changes are hallmarks of many human cancers and lead to concomitant gene inactivation [ 6 8 ]. DNA methyltransferase 1 (DNMT1) is a major DNA methyltransferase responsible for methylation maintenance during DNA replication, and inactivation of DNMT1 in mice results in early embryonic lethality [ 9 ].…”

“…Given that hypoxia is associated with increased malignancy, resistance to therapy and distant metastasis in solid tumors, including NPC [ 20 , 21 ], we hypothesized that hypoxia may mediate the up-regulation of miR-182-5p in NPC. To this end, miR-182-5p levels were measured by qPCR in cells cultured in the hypoxic incubator at different time points.…”

“…V-ATPase and EGFR can antagonize one another [ 32 ]. The induction of EGFR under hypoxic conditions has been found to promote cell proliferation and migration, and patients with hypoxic breast tumors and EGFR hypomethylation may benefit from EGFR inhibition [ 33 ]. Unfortunately, the specific mode of interaction between EGFR and ATP6AP1 in BC is still unknown.…”
