Hypoxia-Inducible Factor1-Α (HIF1α) and Vascular Endothelial Growth Factor-A (VEGF-A) Expression in De Novo AML Patients

Abstract: Background:Bone marrow hypoxia can promote leukemia progression in human cases of acute myeloid leukemia (AML). In addition, low oxygen tension is able to regulate the expression of different genes involved in malignancy. In this study, we hypoxia-inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF-A) genes were assessed as principal regulators of hypoxia in do novo AML patients. Methods:Peripheral blood and bone marrow samples were collected from 57 AML patients and 17 normal control subj… Show more

“…Statistical significance was determined via one-way ANOVA, two-way ANOVA, Tukey's multiple comparisons test or unpaired Student's t-test. Pearson's χ 2 test was used to analyze the relationships between PD-L1 and glycolysis-associated genes.…”

“…Acute myeloid leukemia (AML) is a complex and common myeloid malignancy, which is accompanied by the abnormal proliferation and differentiation of bone marrow precursor cells in the hematopoietic system (1,2). At the present stage, older adults with this disease have a worse prognosis (3).…”

“…In addition to solid tumors, VEGF-A and PlGF play a relevant role in angiogenesis occurring within the bone marrow microenvironment and in acute leukemia spreading at the central nervous system (CNS) site [265][266][267][268][269]. Acute myeloid leukemia (AML) is the most common acute form diagnosed in adults and is characterized by an increased number of immature myeloid cells in the bone marrow, whereas acute lymphocytic leukemia (ALL) is a malignancy of immature lymphocytes that represents 60-74% of all leukemia cases in patients under the age of 20 [270].…”

“…Notably, a correlation between the hypoxia marker HIF1α and VEGF-A has been recently reported. Both are overexpressed in AML cells, but the concentration of HIF1α was significantly higher in AML-M3-derived cells than in other AML cells [ 156 ]. This could be explained by the oncogenic mutation PML-RARA in promyelocytic leukemia (M3) [ 157 ].…”
