ArabidopsisCytochrome P450 Monooxygenase 71A13 Catalyzes the Conversion of Indole-3-Acetaldoxime in Camalexin Synthesis

Abstract: Camalexin (3-thiazol-2-yl-indole) is an indole alkaloid phytoalexin produced by Arabidopsis thaliana that is thought to be important for resistance to necrotrophic fungal pathogens, such as Alternaria brassicicola and Botrytis cinerea. It is produced from Trp, which is converted to indole acetaldoxime (IAOx) by the action of cytochrome P450 monooxygenases CYP79B2 and CYP79B3. The remaining biosynthetic steps are unknown except for the last step, which is conversion of dihydrocamalexic acid to camalexin by CYP7… Show more

“…However, in cat2 sid2 plants, this induction was abolished. A similar expression pattern was observed for two CYP450s involved in camalexin synthesis (Glawischnig, 2007;Nafisi et al, 2007) as well as for camalexin itself and both free and total scopoletin (Fig. 5, C-G).…”

“…2E) were among the 54 genes differentially expressed in the three mutants. In addition, three enzymes implicated in synthesis of the phytoalexin camalexin (PAD3, CYP71A13, and CYP79B2; Nafisi et al, 2007), and three genes involved in the synthesis or maintenance of SA (ICS1, EDS1, and PAD4) are shared by mekk1 and mpk4 and are also up-regulated in mkk1/2, although they are not among the 500 most significantly differentially regulated genes in mkk1/2. Gene ontology analysis of the overlapping genes in mkk1/2, mekk1, and mpk4 revealed SAR to be the most significantly overrepresented term among many other terms implicated in defense response (Supplemental Table S4).…”

“…However, compounds of that class remained undetectable in avocado tissue, hence the function of CYP71A1 in the plant and the physiological substrate are still unclear [30]. CYP71A13, a member of the largest and most diverse family in Arabidopsis, was shown to be involved in the formation of the indole alkaloid specialized metabolite camalexin [31], suggesting that CYP71A family members have acquired highly specific functions across and within individual species. The elusive P450 with in planta geraniol hydroxylase activity was finally discovered in C. roseus, where CYP76B6 was shown to be involved in the route to monoterpenoid indole alkaloids [32].…”
